Investigating Catalase Activity
Study the kinetics of hydrogen peroxide breakdown.
Procedure
Aim
To investigate the effects of temperature, pH, and substrate concentration on the rate of reaction catalyzed by catalase from liver extract.
Step-by-Step Procedure
- 1
Set the desired temperature using the water bath control.
- 2
Adjust the pH of the reaction mixture using the buffer slider.
- 3
Prepare the specified concentration of hydrogen peroxide (substrate).
- 4
Add the catalase source (liver extract) to the substrate and immediately start the data collection.
- 5
Record the volume of oxygen gas produced every 3 seconds for 60 seconds.
- 6
Calculate the initial rate of reaction from the slope of the graph.
